Isochrysis galbana의 냉동보존 효과를 조사하기 위해 해동 후 생존율을 관찰한 결과 각 동해방지제의 1.5 M과 2 M에서 80% 이상의 생존율을 보여 동해방지제 간의 유의한 차이는 없었다. 또한 1 M, 2.5 M, 3 M 실험구에서 60%이상의 생존율을 보여 I. galbana의 경우 동해방지제 농도에 따라 생존율의 차이를 보였다. 냉동보존 후 세포 증식을 조사한 결과 dimethyl sulfoxide의 경우 1.5 M에서 실험개시시 30 × 104 cell/ml이던 것이 3 일 후 107 × 104 cell/ml였고, 실험종료시인 5 일 후에는 365 × 104 cell/ml로 증식되어 가장 높은 증식률을 보였다. Ethylene glycol의 경우 1.5 M에서 실험개시시 30 × 104 cell/ml이던 것이 3 일 후 96 × 104 cell/ml였고, 실험종료시인 5 일 후에는 298 × 104 cell/ml로 증식되어 가장 높은 증식률을 보였다. Glycerol은 1.5 M에서 실험개시시 30 × 104 cell/ml이던 것이 3 일 후 190 × 104 cell/ml, 실험종료시인 5일 후에는 512 × 104 cell/ml로 증식되어 전 실험구 중에서 가장 높은 생존율 및 증식률을 보였다. 1.2-propanedial의 경우 1.5 M에서 실험개시시 30 × 104 cell/ml이던 것이 3 일 후 138 × 104 cell/ml였고, 실험종료시인 5 일 후에는 385 × 104 cell/ml로 증식되어 가장 높은 증식률을 보였다. 참굴 D상 유생을 대상으로 먹이효과를 조사한 결과 실험구와 대조구간 유생의 성장 및 생존율에 유의한 차이를 보이지 않았다.
This study was examined the possibility on the dietary value and cryopreservation of the marine microalga, Isochrysis galbana. Four cryoprotectants (CPAs), dimethyl sulfoxide (DMSO), ethylene glycol (EG), glycerol (Gly) and 1.2-propanedial (PD) were tested at the concentrations of 1.0, 1.5, 2.0, 2.5 and 3.0 M. The highest survival rates were obtained with 1.5 and 2.0 M four cryoprotectants yielding a survival rate of 80%. The result showed that cell reproduction was 30 × 104 cell/ml at the initial point of the experiment and 365 × 104 cell/ml after 5 days in 1.5 M of dimethyl sulfoxide, as well as cell reproduction was 30 × 104 cell/ml at the initial point of the experiment and 298 × 104 cell/ml after 5 days in 1.5 M of ethylene glycol. In addition, cell reproduction was 30 × 104 cell/ml at the initial point of the experiment and 512 × 104 cell/ml after 5 days in 1.5 M of glycerol, as well as cell reproduction was 30 × 104 cell/ml at the initial point of the experiment and 385 × 104 cell/ml after 5 days in 1.5 M of 1.2-propanedial.
