유류 오염지역의 수산물 중 다환방향족탄화수소류 (PAHs) 분석 및 위해평가

초록

허베이 스피리트호 유류유출 사고 후 지역주민들의 수산물 섭취로 인한 건강 위해도를 평가하기 위하여, 가스크로마토그래피-질량분석기(GC-MSD)를 이용하여 유류유출 지역 주민들이 직접 채취 및구입한 수산물 중 다환방향족탄화수소류(PAHs) 16종을 분석하였다. 시료는 KOH로 가수분해하여Methylene chloride로 추출하였다. 추출액 중의 16개의 PAHs는 실리카/플로리실 정제컬럼을 이용하여Methylene chloride : n-헥산(1:9) 혼합용액으로 용출하였고 GC-MSD의 SIM을 이용하여 분석하였다. 16개의 PAHs의 평균 회수율은 79~85%이었으며, 126개의 시료에서 0.17~6.04 μg/kg 수준으로 검출되었고,Benzo(a)pyrene의 독성등가계수(TEF)를 적용한 Benzo(a)pyrene 독성등가량(TEQBaP)은 0~0.91 μgTEQ/kg 수준이었다. 또한 유류유출 지역 주민들의 벤조피렌의 일일 평균 노출량은 5.5×10−8 mg/kg bw/day 이었고, PAHs 일일 평균 만성노출량은 1.3×10−5 mgTEQ/kg bw/day이었다. 그리고 안전마진(MOE)과 초과발암위해도는 각각 1.8×106, 9.8×10−8으로 사람에게 위해영향발생 우려가 낮은 수준이었다.

Abstract

To assess health risk for the intake among residents after the Hebei Spirit oil spill, 16 Polycyclic Aromatic Hydrocarbons (PAHs) in seafood samples from oil contaminated bay were determined by Gas Chromatography-Mass Spectrometry (GC-MSD) and samples were personally collected and purchased by residents. Samples were hydrolyzed with KOH and extracted with methylene chloride. The extracted solution were cleaned up using silica/florisil column and 16 PAHs were eluted by methylene chloride : n-hexane (1:9)mixture and determined by GC-MSD in Selected Ion Monitoring (SIM) mode. The mean recoveries for 16PAHs ranged from 79% to 85%. The 16 PAHs levels in 126 samples ranged from 0.17 to 6.04 μg/kg and the TEQBaP (Toxic EQuivalents) levels in 126 samples were calculated using benzo(a)pyrene toxic equivalency factor for individual 16 PAHs and ranged from 0 to 0.91 μgTEQ/kg. The average Benzo(a)pyrene dietary exposure of residents was 5.5×10−8 mg/kg bw/day and the average PAHs chronic dietary exposure was 1.3×10−5mg TEQ/kg bw/day. The margin of exposure (MOE) and the excess cancer risk and were 1.8×106 and 9.8×10−8,respectively. Therefore, the assessment result was considered as low concern for health risk.
